I seriously doubt they'd fill the new one yet, because its a control drug. You will probably have to take the new amount and use the old pills until they are used up, then get the new one filled.
If the quantity changes, it’s considered a new script entirely and legally then can be picked up as soon as it’s filled if the doctor doesn’t designate a “fill date”. So as far as the computers go, even though it’s a controlled substance, the computer sees a new script with new directions and quantity and wont affiliate it with the other so the only way he wouldn’t be able to put it up immediately is if the doctor puts a note to the pharmacist, or, some pharmacists will take it upon themselves to call the doctor and report that he just picked one up, suggesting there be a wait and in that case the doctor can say it’s fine or listen to the pharmacists suggestion. If the doctor and pharmacist don’t intervene though, the changed quantity officially makes it a separate script entirely from the first and therefore available immediately.
