Moderate Potential Hazard, Moderate plausibility. Applicable conditions: History - Musculoskeletal Disorder

Musculoskeletal adverse reactions, which may be accompanied by serum creatine kinase elevations have been reported with the use of drugs that inhibit the hedgehog pathway. It is recommended to obtain serum creatine kinase levels and renal function tests prior to initiating therapy, periodically during treatment, and as clinically indicated. Care should be taken when prescribing these agents to patients presenting musculoskeletal pain or myalgia. Temporary dose interruption or discontinuation of therapy may be required based on the severity of musculoskeletal adverse reactions.

Moderate Potential Hazard, Moderate plausibility. Applicable conditions: Renal Dysfunction

Mild (CrCl 60 to 89 mL/min) and moderate (CrCl 30 to 59 mL/min) renal impairment had no effect on sonidegib steady-state exposure. Caution is recommended when this agent is administered to patients with severe renal impairment as no data is available from the manufacturer.
