Jakafi (ruxolitinib) for Myelofibrosis "Been on it for the past 5 years, starting with 5 mg 2x a day and now 25 mg 2x a day, which is throwing off my hemoglobin and lowering platelets. Enlarged spleen 23 cm lets me know when it's time to up the dosage. I have gained over 20 pounds, and a lot of it is swelling, looking pregnant, but I'm a dude. This past week I had to stop taking it for two days because my hemoglobin/platelet count dropped too low (I had to stop shaving and do no type of manual labor). The side effects were tough: severe migraines - had to black out my room, dizzy, faint, nausea, balance/coordination issues, brain fog, laying in bed for most of 48 hours. I had to go back on it because my spleen was exploding in size and hurting really bad. I need to get off of it and find a newer drug or clinical studies."

Jakafi (ruxolitinib) for Graft-versus-host disease "Taking Jakafi for 1 year 3 months, working well with no side effects. Take 5 mg daily. I take it for GvHD of the skin, and it has done a great job controlling it. I have had no flare-ups."

For Graft-versus-host disease "Works great. Hardly any side effects. Had mostly mouth cgvhd after transplant 4 years ago. This helped me to greatly reduce prednisolone."

For Myelofibrosis "I have been on Ruxolitinib for about 9 months now. I started at 40mg/day and am down to 15mg/day now. My RBC has deteriorated significantly (even at low dosage), which has led to some significant fatigue. I was on Hydroxyurea for 10 years prior to this until I started to experience some uncomfortable side affects (quite painful Neuropathy), night sweats etc. I am quickly coming to the conclusion that this drug is not for me. With haemoglobin levels less than half of normal, it is hard to function properly. My Doctor just started me (6 weeks) on Eprex to try and stimulate more red blood cells but the results have not been positive so far. I am becoming somewhat despondent and not sure where to turn for an alternative course of treatment. Ruxolitinib is just not working."
