No, when you take Xifaxan with a probiotic, neither cancels out the effect of the other, although you may get the most benefit from both by separating the times at which you take them as much as you conveniently can. Xifaxan is a broad-spectrum antibiotic, but it does not wipe out all of the microbiome in your gut and in fact leaves it substantially unchanged overall.
